Abstract

Damnacanthal, een antrachinon verbinding die wordt geïsoleerd uit de wortels van Morinda citrifolia L. (noni), wordt gebruikt voor traditionele therapie bij een aantal chronische ziekten waaronder kanker. Hoewel noni al lange tijd wordt gebruikt in de Aziatische en Polynesische landen, krijgen de moleculaire mechanismen die verschillende voordelen geven, nu meer bekendheid.. In dit rapport hebben we  systematische onderzocht wat de werking is op de kanker-onderdrukkende vermogen van damnacanthal in colorectale tumorvorming. Damnacanthal vertoont en stop van de celgroei evenals caspase activiteit ( dit zijn protease die aanzetten tot apoptose = celdood) vergroting in colorectale kanker cellen. We hebben ook verschillende targetproteine onderzocht en vonden dat het pro-apoptotische eiwit steroïde anti-inflammatoire geactiveerde gen-1 (NAG-1) wordt sterk geïnduceerd (op gang gebracht). Vervolgens hebben wij ook gevonden dat damnacanthal verbetering brengt in de transcriptiefactor CCAAT / versterker bindend eiwit β (C / EBPB), die NAG-1 transcriptionele activiteit regelt. Blokkering van C / EBPB van shRNA resulteert in de vermindering van NAG-1 expressie en caspase activiteit in aanwezigheid van damnacanthal. Tezamen geven deze resultaten aan dat damnacanthal een verhoging geeft van antitumorigene activiteit in humane colorectale kankercellen en C / EBPB speelt een rol in damnacanthal geïnduceerde NAG-1 expressie.

Damnacanthal, a noni component, exhibits antitumorigenic activity in human colorectal cancer cells.

Abstract

Damnacanthal, an anthraquinone compound, is isolated from the roots of Morinda citrifolia L. (noni), which has been used for traditional therapy in several chronic diseases including cancer. Although noni has been consumed for a long time in Asian and Polynesian countries, the molecular mechanisms by which it exerts several benefits are starting to emerge. In this report, we examined systematic approaches on the cancer-suppressing capability of damnacanthal in colorectal tumorigenesis. Damnacanthal exhibits cell growth arrest as well as caspase activity induction in colorectal cancer cells. We also examined several potential target proteins and found that the proapoptotic protein nonsteroidal anti-inflammatory activated gene-1 (NAG-1) is highly induced. Subsequently, we have found that damnacanthal also enhances transcription factor CCAAT/enhancer binding protein β (C/EBPβ), which controls NAG-1 transcriptional activity. Blocking of C/EBPβ by shRNA results in the reduction of NAG-1 expression as well as caspase activity in the presence of damnacanthal. Taken together, these results indicate that damnacanthal increases antitumorigenic activity in human colorectal cancer cells and that C/EBPβ plays a role in damnacanthal-induced NAG-1 expression.
